CYTOSINE ARABINOSIDE (Cytarabine)

It is phosphorylated in the body to corresponding nucleotide, which inhibits DNA synthesis. It also interferes with DNA repair. Cytorabine is cell cycle specific and acts primarly during S phase. Its main use is to induce remission in acute leukemia.

Indications

Acute leukaemias, lymphosarcomas.


Dosage

3-6 mg/kg body wt i.v daily in 2 divided doses for 5-10 days.


Contra-Indications

Hypersensitivity, pregnancy.


Special Precautions

Hepatic impairment. Paediatrics: Indicated in certain leukaemia, under strict supervision. Pregnancy: Safety not established Lactation: Safety not established Elderly: Use with caution.


Side Effects

Bone marrow suppression. G.I.T disturbances.


Drug Interactions

Potentiates bone marrow depression with radiotherapy and other myelotoxic drugs. Reduces plasma concentratrion of digoxin, antagonises gentamycin. Decreased efficacy of flurocytosine.
