Protamine sulphate

Protamine Sulfate Injection, USP is a sterile, non-pyrogenic, isotonic solution of protamine sulfate in Water for Injection. It acts as a heparin antagonist. It is also a weak anticoagulant.

Indications

Antidote for heparin in uncontrolled bleeding.


Dosage

1 ml of protamine sulphate neutralises the effect of 1000 i.u. of circulating heparin.


Contra-Indications

Protamine sulfate is contraindicated in patients who have shown previous intolerance to the drug.


Special Precautions

Avoid rapid administration. Fish allergy. Previous exposure to protamine (including insulin). Severe left ventricular dysfunction. Abnormal pulmonary hemodynamics. Previous vasectomy. Pregnancy (Cat.C). Nursing mothers.


Side Effects

Hypertension, nausea, vomiting, lassitude. Back pain, blood pressure, bradycardia, dyspnea, or transitory flushing and a feeling of warmth


Drug Interactions

Protamine sulfate has been shown to be incompatible with certain antibiotics, including several of the cephalosporins and penicillins
