Bayer Bets Big on Parkinson's, Advancing Both Cell and Gene Therapies in Global Trials
Bayer, the pharmaceutical giant, has revealed that it is the first business to move both a cell therapy and a gene therapy for Parkinson's disease into mid- and late-stage clinical trials at the same time, which marks an important turn in neurodegenerative disease research. This dual-track approach represents a significant industry push towards creating medicines that have the potential to change the course of the disease rather than simply managing its symptoms
Bayer announced the start of its key Phase III trial, exPDite-2, for the cell therapy bemdaneprocel, with the first participant receiving treatment. Concurrently, the Phase II trial for its gene therapy, AB-1005, known as REGENERATE-PD, began randomising participants in the United Kingdom and Poland, with Germany and the United States following. Both medicines target moderate stages of Parkinson's disease
Parkinson's disease, which affects well over 10 million people worldwide and is the fastest-growing neurodegenerative condition, is defined by the gradual loss of dopamine-producing neurones in the brain. This causes severe motor symptoms such as tremors and rigidity. Current treatments essentially replace dopamine, but they lose efficacy over time and do not reduce disease progression
Bayer's two investigational medicines approach the problem from distinct perspectives. The cell therapy, bemdaneprocel, works as a "cellular replacement" technique. It entails surgically introducing dopamine-producing neurone precursors produced from stem cells into the brain. These cells are intended to develop and re-establish the neuronal networks damaged by Parkinson's.
"Bemdaneprocel aims to replace these lost cells with new dopamine-producing neurones," said Dr Gabi Belfort of BlueRock Therapeutics, a Bayer subsidiary developing the treatment
In contrast, the gene therapy, AB-1005, is a "neuroprotective" strategy. It uses a harmless virus to deliver a gene known as GDNF straight to the brain. This gene instructs brain cells to continuously create a protein that acts as fertiliser for dopaminergic neurones, potentially extending their survival and functionality
This combined method increases the likelihood of a breakthrough while acknowledging Parkinson's complexity. "Our dual approach exemplifies our strategic vision and maximises our chances of offering renewed hope for Parkinson's patients who have been waiting for new therapies for too long," stated Christian Rommel, Head of R&D at Bayer Pharmaceutical Division
For patients, the appeal of these one-time surgical treatments is the potential for long-term benefit, which contrasts sharply with the existing requirement for several daily drugs. The emphasis on moderate-stage disease is critical; acting at this stage may preserve function before irreversible damage develops
Despite the optimistic rationale, Bayer emphasises that both medicines are still experimental and have not yet been approved by any regulatory authorities. Their efficacy and safety are still being determined. The Phase III trial for bemdaneprocel will compare it to a fake surgery, which is a strict method needed to show real benefits
The trials' global scope, which includes Europe and North America, is particularly notable. It speeds up recruitment and ensures that the results apply to a varied patient group. As the world's population ages, there is an increasing need for Parkinson's disease-modifying medicines. Bayer's significant bet marks a watershed moment in the protracted battle to alter the course of this deadly disease.
