Danozol

It is an orally active impeded testosterone derivative. It has mild androgenic, anabolic and progestational activity. However, the most prominent action is suppression of gonadotrophin secretion frm pituitary both in men & women leading to inhibition of testicular/ovarian function.

Indications

Endometriosis. Menorrhagia. Fibro- cystic breast disease. Gynaecomastia. Precocious puberty in boys. Infertility due to endometriosis.


Dosage

Endometriosis & associated infertility : 200-400 mg daily for 3-4 mths. Fibrocystic breast disease & gynaecomastia : 100-400 mg daily for 2-3 mths. Menorrhagia 200 mg daily for 12 wks.


Contra-Indications

Pregnancy, abnormal genital bleeding of unknown etiology, lactation. Porphyria. Renal or cardiac oedema.


Special Precautions

Severe hypertension, Diabetes. History of jaudice due to oral contraceptives. Epilepsy, migraine, cardiac hepatic or renal dysfunction. Weight gain. Monitor liver function tests.


Side Effects

Complete amenorrhoea, occasional spotting. Flushes, night sweats, acne, hirsutism, weight gain, G.I. upsets. Loss of libido & muscle cramps.


Drug Interactions

Increased incidence of insulin resistance. Potentiates action of oral anticoagulants. Oral contraceptive interfere with danazol action. Adjustment in antiepileptics dose required.
