Lamivudine with Zidovudine

Lamivudine with Zidovudine is a combination antiretroviral medication used in the treatment of HIV (Human Immunodeficiency Virus) infection. Lamivudine belongs to the nucleoside reverse transcriptase inhibitor (NRTI) class, and Zidovudine is also an NRTI. Together, they work to inhibit the replication of the virus.

Indications

-HIV Infection: Used as part of combination therapy to manage HIV infection in adults and pediatric patients. -Treatment of HIV infection.


Dosage

-Dosage is determined based on the patient's weight, age, and specific HIV treatment guidelines. The combination is typically administered orally, with or without food. -Adults and adolescents (at least 12 years of age) is one tablet (containing 150mg of lamivudine and 300mg of zidovudine) twice daily. It should not be prescribed for patients requiring dosage adjustment such as those with reduced renal function (creatinine clearance <50 mL/min), those with low body weight (<50 kg or 110lb), or those experiencing dose-limiting adverse events.


Contra-Indications

-Hypersensitivity: Avoid in patients with known hypersensitivity to lamivudine, zidovudine, or other components of the combination. -Severe Anemia: Contraindicated in patients with severe anemia due to zidovudine.


Special Precautions

-Renal Impairment: Adjustments in dosage may be necessary in patients with renal dysfunction. -Hematologic Monitoring: Regular monitoring of blood counts is recommended, especially in patients with a history of bone marrow disorders.


Side Effects

Common side effects may include nausea, headache, and fatigue. Serious side effects are rare but may include lactic acidosis, hepatomegaly, and hematologic abnormalities.


Drug Interactions

-Didanosine: Administer lamivudine and zidovudine at least one hour before or after didanosine. -Ribavirin: Potential for increased risk of anemia when used with zidovudine.
