PACLITAXEL

PACLITAXEL

It is a diterpenoid compound with anti-tumour activity. It binds specifically to the b-tubulin subunit of microtubule and appears to antagonise the disassembly of this cytoskeletal protein.

Indications

Metastatic ovarian and breast cancer.


Dosage

175 mg per square metre by iv infusion over 3 rs. Repeat every 3 weeks. Pretreat with corticosteroids,antihistaminics and H2 antagonists.


Contra-Indications

Hypersensitivity.


Special Precautions

Monitor haematological functions. Paediatrics: Not recommended. Pregnancy: Not usually prescribed. Lactation: Not advised.


Side Effects

Bone marrow depression, hypersensitivity reactions, myalgia, chest pain, bradycardia, a stocking-glove sensory neuropathy, mucositis, nausea, vomiting, diarrhoea, alopeia.
