Efonidipine

Efonidipine is a calcium channel blocker that is used for the management of hypertension (high blood pressure). Efonidipine is a dihydropyridine calcium channel blocker which blocks both T-type and L-type calcium channels. Efonidipine is a dihydropyridine calcium channel blocker that acts on the L-type calcium channels in vascular smooth muscle cells. It inhibits the influx of calcium ions, leading to vasodilation and a reduction in blood pressure.

Indications

-Efonidipine is indicated for the treatment of essential hypertension (high blood pressure) in adults. -For the treatment of essential hypertension, renal parenchymal hypertension and angina.


Dosage

-The recommended dosage of efonidipine may vary depending on individual patient factors. Typically, the starting dose is 20 mg once daily, and it can be titrated up to 40 mg once daily if necessary. -oral:20/40 mg tablet;20-80 mg per day.


Contra-Indications

-Efonidipine is contraindicated in patients with a known hypersensitivity to the drug or its components. It is also contraindicated in patients with severe hepatic impairment. -hypersensitivity to efonidipine.


Special Precautions

-Caution is advised in patients with impaired renal function. -Efonidipine should be used with caution in patients with congestive heart failure. -Patients should be monitored for hypotension, especially during initial titration. -Use with caution in case of hepatic impairment or SA node dysfunsion. Paediatric: safety and efficacy of in pediatric patients have not been established. Pregnancy: Data no sufficient. Lactation: Data no sufficient. Elderly: Use with caution, increase change of hypotension.


Side Effects

-Common side effects of efonidipine may include peripheral edema, headache, dizziness, flushing, and palpitations. Less common side effects may include constipation, nausea, and fatigue. -Hot flushes, palpitation, facial flushing and headache, Frequenturination, pedal edema, elevation in serum total cholesterol, ALT (SGPT), AST (SGOT) and BUN may occur.


Drug Interactions

-Efonidipine may interact with other antihypertensive medications, leading to an additive effect. It is important to monitor blood pressure regularly and adjust doses accordingly. Additionally, caution is advised when co-administering efonidipine with strong CYP3A4 inhibitors or inducers. -other antihypertensive drugs, cimetidine, grape fruit juice, tacrolimus.
